Se diseñó y construyó un Reactor Anaerobio de Lecho Fluidizado – RALF – a escala piloto, para tratar un efluente similar a las "aguas madres" – provenientes del procesamiento de aserrín de quebracho colorado detanizado – de una industria taninera. Las aguas madres poseen: acidez acentuada, elevada carga orgánica y compuestos orgánicos complejos; principalmente, ácido acético y furfural que la tornan un efluente recalcitrante. Actualmente, éste es tratado en la fábrica con un sistema de Pantanos Secos Artificiales – PSA – que utiliza fitoextracción para eliminar contaminantes. Para remover el furfural, se evaluó la aclimatación de un consorcio bacteriano autóctono – extraído de lodos de los sedimentadores del PSA – en un medio mineral M9 con distintas concentraciones de furfural. Se incubó el medio en condiciones de oxígeno reducido y en estufa a 37ºC durante 7 días; se comprobó el crecimiento bacteriano visualmente. Se monitoreó el desempeño del RALF, operando con el consorcio aclimatado, analizando los porcentajes de remoción logrados. Los resultados demostraron que se pueden adaptar bacterias hasta 630 mg/L de furfural en aguas madres. La eficiencia de remoción de furfural, durante 20 h de funcionamiento del reactor, fue del 99%.
An Anaerobic Fluidized Bed Reactor – RALF – was designed and built at pilot scale to treat an effluent similar to the "mother liquor" – originated from the processing of detannized red quebracho sawdust - from a tanning industry. The mother liquor has: elevated acidity, high organic content and complex organic compounds; mainly, acetic acid and furfural that become a recalcitrant effluent. Currently, it is treated at the factory with a system of Artificial Dry Wetlands – PSA – which uses phytoextraction to remove contaminants. In order to remove furfural, the acclimatization of an autochthonous bacterial consortium - extracted from sludge from the RALF settlers - was evaluated in an M9 mineral medium with different concentrations of furfural. The medium was incubated under reduced oxygen content and in an oven at 37 ° C for 7 days; bacterial growth was visually checked. The performance of the RALF was monitored, operating with the acclimatized consortium, analyzing the percentages of removal achieved. The results showed that bacteria can be adapted up to 630 mg/L of furfural in mother liquor. The removal efficiency of furfural, for 20 h of the reactor operation, was 99%.

El calentamiento global debido al aumento de las emisiones de gases que producen efecto invernadero es uno de los problemas más importantes a ser abordados en nuestros días. El dióxido de carbono es el principal gas que contribuye al efecto invernadero y cuya concentración está aumentando bruscamente. Para combatir esta severa problemática, han sido propuestas numerosas tecnologías destinadas a su captura y almacenamiento. En este sentido, los ecosistemas forestales son considerados grandes sumideros terrestres de gases de efecto invernadero (GEI) pues absorben CO2 atmosférico, almacenándolo en la biomasa y fijándolo al suelo. Los hongos micorrízicos arbusculares (HMA) producen una glicoproteína (glomalina) que acumula carbono directamente a través del carbono contenido en la propia molécula. Además actúa como material cementante, interviniendo directamente en la formación de los agregados del suelo. La glomalina se encuentra en las hifas de los HMA y es liberada al suelo mediante el proceso de descomposición de las mismas. El objetivo de este estudio fue cuantificar y comparar el contenido de glomalina total y fácilmente extraíble en tres áreas de estudio: bosque implantado de 20 años de edad con la especie Pino radiata (Pinus radiata D. Don), bosque implantado de 50 años de edad con la misma especie y un área de bosque nativo cuya especie predominante es Ciprés de la Cordillera (Astrocedrus chilensis). Los niveles de glomalina se relacionaron con el contenido de materia orgánica y con la actividad microbiológica del suelo. Los resultados mostraron los mayores niveles de glomalina total y fácilmente extraíble en el suelo debajo de Pino radiata de más de 50 años de implantación 16,8 y 4,13 mg.g-1, respectivamente. Para la plantación de Pino de 20 años los valores fueron 10,7 y 2,80 mg.g-1 y para el bosque nativo 9,0 y 1,67 mg.g-1. Se hallaron los mayores valores de materia orgánica y actividad microbiológica para al bosque nativo y el bosque de Pino de más de 50 años. No se hallaron diferencias significativas en la actividad microbiológica medida en el suelo de bosque nativo respecto del suelo con la plantación de 50 años. Conclusiones: la especie Pino radiata sería importante como especie secuestrante de carbono, observándose altos contenidos de glomalina aún en plantaciones de 20 años de edad, donde los valores obtenidos para la glomalina ya superan al bosque nativo. Los valores más altos de materia orgánica encontrados en el bosque implantado de 50 años correlacionaron positivamente con los contenidos de glomalina, debido a que esta glicoproteína participaría en el recubrimiento de los residuos orgánicos impidiendo su rápida degradación. Por lo tanto este trabajo da indicios para seguir realizando investigaciones que corroboren que plantaciones con Pino radiata serían una alternativa para mitigar emisiones de CO2 a la atmósfera.
Global warming, otherwise known as climate change, is a major subject that needs to be addressed. This is due to the increasing concentration of greenhouse gas emissions, primarily carbon dioxide (CO2). Numerous technologies have been proposed for their capture and storage. In this sense, forest ecosystems are considered large terrestrial sinks of greenhouse gases (GHG) due to absorbing atmospheric CO2 and storing it in the biomass and fixing it to the soil. Arbuscular mycorrhizal fungi (HMA) produce a glycoprotein (glomalin) that accumulates carbon directly in the molecule itself. It also acts as a cementing material, intervening directly in the formation of soil aggregates. The glomalina is found in the hyphae of the HMA and is released to the soil by decomposition of those hyphae. The aim of this study was to quantify and compare the total glomalin and easily extractable glomalin content in three study areas; 20-year-old pine forest with Pinus radiata (Pinus radiata D. Don), 50-year-old implanted forest with the same species, and one area of native forest whose predominant specie is the Cypress (Austrocedrus chilensis). The levels of glomalin were related to the content of organic matter and to the microbiological activity of the soil. The results showed the highest levels of total glomalin and easily extractable glomalin in the soil under Pinus radiata of more than 50 years of implantation 16,8 and 4,13 mg.g-1, respectively. For the 20-year-old pine plantation the values were 10.7 and 2.80 mg.g-1 and for the native forest 9.0 and 1.67 mg.g-1. For the native forest and the Pine- radiate, the highest values of organic matter and microbiological activity were more than 50 years. No significant differences were found in the microbiological activity measured in native forest soil compared to the soil with Pinus radiata 50 year of plantation. In conclusion, the Pinus radiata is as important as carbon sequestering, evident in the high glomalin content in the soil observed even in plantations of 20 years, where the values obtained for glomalina already exceeded the values obtained in the native forest of Austrocedrus chilensis. The highest values of organic matter found in the Pinus radiata implanted of 50 years correlated positively with the content of glomalina. This is because the glycoprotein would participate in the coating of the organic residues, preventing its rapid degradation. Therefore, this work suggests to continue conducting research that corroborate that plantations with Pinus radiata would be an alternative to mitigate emissions of CO2 to the atmosphere.

Las bacterias del género Bacillus poseen una reconocida actividad como promotoras del crecimiento vegetal. Dependiendo de las condiciones de cultivo, las células de B. subtilis cambian de tener un crecimiento de vida libre como células planctónicas, a convertirse en células no móviles para formar una biopelícula. Por otra parte, en condiciones de limitación de nutrientes o en respuesta a condiciones ambientales adversas las células de Bacillus detienen su normal proceso de división celular para dar inicio a otro proceso altamente organizado que culmina con la producción de una espora muy resistente. Los objetivos de este trabajo fueron: 1-Optimizar las condiciones de cultivo para el desarrollo de una biopelícula robusta y su vinculación con el proceso de esporulación. 2- Evaluar el efecto de la inoculación de semillas de Lactuca sativa var. Crimor cuando Bacillus subtilis fue aplicado en su estado planctónico y bajo la forma de biopelícula. Se utilizó Bacillus subtilis subsp. spizizenii crecida en medio salino suplementado con distintas fuentes carbonadas simples, aminoácidos y bajo distintas velocidades de agitación. Los recuentos bacterianos se realizaron a través de diluciones seriadas y siembra en placas con agar nutritivo. Para el ensayo de inoculación, el crecimiento bacteriano fue sin agitación para el caso de la biopelícula y a 150 rpm para el inóculo líquido (estado planctónico). Las semillas de Lactuca sativa var. Crimor fueron sembradas e inoculadas con un cultivo líquido de la bacteria (en su forma de vida planctónica) mientras que en otras semillas se depositó asépticamente una fina porción de la biopelícula. Se preparó para la siembra una mezcla de tierra y compost en proporción 3:1 (sustrato). Se realizaron tratamientos utilizando esta mezcla tindalizada (sustrato empobrecido) y sin tindalizar (sustrato fértil). Los resultados mostraron que las fuentes carbonadas más eficaces para la formación de la biopelícula fueron glicerol, glucosa y manitol, siendo la primera la más resistente a los procesos degradativos. La fuente nitrogenada óptima fue ácido L-glutámico. La biopelícula solo pudo desarrollarse en condiciones estáticas o cuando la agitación fue baja (80rpm). No se detectaron diferencias en el número de esporas liberadas en presencia o ausencia de biopelícula. Bacillus actuó como promotor del crecimiento sobre Lactuca sativa var. Crimor, resultando efectiva tanto cuando se la aplicó en su estado planctónico como de biopelícula. El efecto promotor del crecimiento vegetal se observó tanto en el sustrato fértil como en el sustrato empobrecido en nutrientes y microorganismos, aunque fue mayor sobre éste último. Así mismo la biopelícula tuvo mayor efecto benéfico como inoculante que la forma planctónica. Estos resultados sugieren que la estructura de la biopelícula le permitiría a la bacteria adaptarse a las nuevas condiciones ambientales, sobre la semilla, favoreciendo la supervivencia y actividad benéfica de la misma sobre la planta. Este trabajo sugiere que la aplicación de biopelículas como inoculante de semillas es una alternativa novedosa para el desarrollo de una agricultura sustentable, que aumente los rindes de los cultivos sin provocar los problemas ambientales vinculados con los fertilizantes químicos.
Bacillus bacteria have a recognized activity as plant growth promoters. Depending on the culture conditions, B. subtilis cells change from having a free-living growth like planktonic cells to non-mobile cells to form a biofilm. On the other hand, under conditions of nutrient limitation or in response to adverse environmental conditions the Bacillus cells stop their normal process of cell division to start another highly organized process that culminates with the production of a very resistant spore. The objectives of this work were: 1-Optimize crop conditions for the development of a robust biofilm and its linkage with the sporulation process. 2- Evaluate the effect of inoculation of seeds of Lactuca sativa var. Crimor when Bacillus subtilis was applied in its planktonic state and in the form of biofilm. Bacillus subtilis subsp. Spizizenii grown in saline medium supplemented with different single carbon sources, amino acids and under different stirring rates. Bacterial counts were performed through serial dilutions and plated on nutrient agar plates. For the inoculation test, the bacterial growth was without agitation for the biofilm case and at 150 rpm for the liquid inoculum (planktonic state). Lactuca sativa seeds of var. Crimor were planted and inoculated with a liquid culture of the bacteria (in their planktonic life form) while in other seeds a fine portion of the biofilm was deposited aseptically. A mixture of soil and compost in a 3: 1 ratio (substrate) was prepared for sowing. Treatments were performed using this blend (immobilized substrate) and without tindalizing (fertile substrate). The results showed that the most efficient carbon sources for biofilm formation were glycerol, glucose and mannitol, the first being the most resistant to the degradation processes. The optimal nitrogen source was L-glutamic acid. The biofilm could only be developed under static conditions or when the agitation was low (80rpm). No differences were detected in the number of spores released in the presence or absence of biofilm. Bacillus acted as a growth promoter on Lactuca sativa var. Crimor, proving effective both when applied in its planktonic state and biofilm. The plant growth promoting effect was observed in both the fertile substrate and the substrate depleted in nutrients and microorganisms, although it was greater on the latter. Also, the biofilm had greater beneficial effect as an inoculant than the planktonic form. These results suggest that the structure of the biofilm would allow the bacteria to adapt to the new environmental conditions, on the seed, favoring the survival and beneficial activity of the same on the plant. This work suggests that the application of biofilms as a seed inoculant is a novel alternative for the development of a sustainable agriculture that increases yields without causing the environmental problems associated with chemical fertilizers.
